Application
CHW Antigen + Ehrlichia Antibody+ Lyme IgG/IgM Antibody Combo Rapid Test Cassette (Whole Blood/Serum/Plasma) is a combined cassette to differentially diagnose the presence of Canine Heartworm antigen and Ehrlichia antibody and Lyme IgG/IgM antibody in canine’s whole blood or serum and plasma.
PRINCIPLE
CHW Antigen + Ehrlichia Antibody + Lyme IgG/IgM Antibody Combo Rapid Test Cassette is based on lateral flow immunochromatographic assay. The test cassette has three testing windows. Each window has an invisible T (test) zone and C (control) zone. When sample is applied into the sample well on the cassette, the liquid will laterally flow on the surface of the test. If there is enough Canine Heartworm antigen or Ehrlichia antibody or Lyme IgG/IgM antibody in the sample, a visible T line will appear in the corresponding testing window. The C line should always appear after a sample is applied, indicating a valid result. By this means, the cassette can accurately indicate the presence of Canine Heartworm antigen or Ehrlichia antibody or Lyme IgG/IgM antibody in the sample.

DIRECTIONS FOR USE
Allow the test cassette, specimen, buffer, and/or controls to equilibrate to room temperature (15-30 °C) prior to testing.
1. Collect canine’s fresh whole blood or serum or plasma samples and ensure that the samples cannot be turbid or precipitated. Only use clear, non-hemolyzed specimens. Do not leave the specimens at room temperature for prolonged periods. Serum and plasma specimens may be stored at 2-8 °C for up to 3 days. For long term storage, serum and plasma specimens should be kept below -20°C
2. Place the test cassette on a clean and level surface.
3. Hold the dropper vertically and transfer 1 drop of sample (approximately 10 μl) and immediately add 3 drops of buffer (approximately 120 μl) into each specimen well (S), then start the timer. See illustration above.
4. Read the result in 10 minutes. Do not interpret results after 20 minutes.
Note: For Ehrlichia window, if the test line is very strong or even darker than the control line or the control line disappears, it may be a strong positive sample other than invalid result. It is recommended to dilute the sample by 3-5 times with the buffer and retest with a new test to confirm the result.
INTERPRETATION OF RESULTS
CHW Ag/Ehrlichia Ab Positive: The presence of both C line and T line
Lyme IgG Positive: Two distinct colored lines appear. One colored line should be in the control region (C) and another colored line should be in the IgG region.
Lyme IgM Positive: Two distinct colored lines appear. One colored line should be in the control region (C) and another colored line should be in the IgM region.
Lyme IgG and IgM Positive: Three distinct colored lines appear. One colored line should be in the control region (C) and another two colored lines should be in the IgG and IgM regions.
Negative: Only clear C line appears.
Invalid: No colored line appears in C zone, no matter whether T line appears. For Ehrlichia window, if the T line is very strong but no C line or weak C line, it may be a result of strong positive sample, it is recommended to dilute the specimen and retest with a new test to confirm.
